rants, eh? Ok.
I was working with my PI yesterday (principal investigator)... and just working with her made me really really nervous and scared to death. After all, she's the PI! 20 times more experience and brains than I have. She was already having a bad day. And I put my samples in the water bath to react for 15 mins and set a timer. However, the timer was only syncronized to the FIRST sample put in the bath. So, when I took all the samples out, the last samples had not been in there a full 15 mins. So, the DNA probably did not replicated and obviously, she was really mad at me. I was trying to explain myself and said "Oh... well...." and she took that as "oh well, I don't care!" Of course I care. I wanna get this right, it's my future! So, today, I'm running agarose gels to check if my DNA did replicate and nothing is coming out in the UV pictures. The good part is that even the DNA that indeed reacted for 15 mins is not showing up, which means we messed something up in the first part of the experiment, which was adding short oligos to our single stranded DNA and she watched me do all that. I'm probably gonna get my head cut off today and I'm really frustrated. On top of that, my FT335 cells overgrew, meaning I probably messed up during plating. But the overgrowth problem was in the 10-1 and the 5x10-2 dilutions, which are basically putting 100 ul and 50 ul in a plate! I couldn't have possibly messed up that. Besides, the other cells I plated in the SAME day, PP102, came out perfect. Which leads me to think FT335 is simply overexpressing the gene that causes it to survive in the FuDR Chlor plates. Hmmm.
Nothing's working. grrr.

Garages, how do they get away with charging so much?? I had to spend £350 on my car and I bet if I'd bought the parts myself it would've been a quarter of that.
And door handles, what are they all about. Yesterday I ripped a shirt on one and tonight I ripped my jeans, bloomin things. I need those swooshy doors from TOS in my house I think.

I can't take it anymore!! Not only has my computer come back from repair speaking german in the scanner instructions, but now it insists I don't have a microsoft virtual machine thing to access my own flippin webpage editor!!!!!!!!!!!! I've been working on the thing for three days straight and it feels like someone took it and shook it and everything's all jumbled up.
I may have to take it up the garden and burn it

I hate being home for more than a weekend. I love my family, but theres a temperment there...Have one little bitty thing go wrong and mum and sister are curing and slamming doors and the what not for all the rest of the day. Makes the other inhabitants miserable.

I love my sister (i guess) b/c well, she's my sisiter, but for the life of me,... I DONT LIKE HER!!!!!!! No consideration fer others whatsover. Im watching a movie on the TV downstairs, she pitches a fit. Hello. there are 3 other TVs in the house!!!!!! Moms making dinner, it doesnt help that we're a bit short on some of the ingredients...dear sister hauls out her smoothie machine cuz she wants one.
She's 23, Ill be 21 in... 20ish days. Unless dear old Dads around, she cant say a civil sentence to me that doesnt inlcude curising at me. Its gotten to the point where you can call me the nastiest names you can think of and I wont blink (can you tell i wasnt liked as a child?). But it DOES get old after awhile...She doesna treat Mum much better...
I wish we could curse..... for my uncensored rants, I have a deadjournal (somebody decided to mock livejournal), my screens DragonPhoenix. ITs a tad outdated, i have to be caredull on this compy.

I hate impatience. Of any kind. Patience is one of the few things I like about me (I have LOTS) and it pisses me off when people get impatient.
We went out to eat, busy place. we were kinda on the other side of the kitchen. Waitress takes our drink order. a few seconds later. one of our party "wheres our drink? Im thirsty" BLOODY HELL!!!!! We're on the OTHER side of the place, and she was flagged down by another patron...SHEESH
I also cant stand that these messages wont stay put if you accidently go to another page. I had a huge list of quizzies already to post on the Quiz and TEsts forum. Then I click on on more test to take...it opens in the same browser window, I hit the back button, low and behold...10 minutes of copy/paste is gone. Screw it.

Oh you know, there are so many things in this world that piss me off. For example: The fact that Nasa, nor anyone else in the space industry can come up with something that will fly at warp, it's not beyond us, that bastards just think it will cost way to much.. Ummm duh! star trek you idoits, lets unite the world, have a close encounter and go to the horsehead nebula! C'mon, Build a few orbital colonies, colonize the moon, WHICH I CAN'T GRASP WHY PEOPLE AREN'T LIVING THERE ALREADY!!! Duh we went there so many years ago and still havent gone back.. C'mon bush! Lets get things going here! I'm not saying screw homeland security, But throw some money at some small coperation willing to buil colonies on the moon! hell I'd live there!!!

you know.. I really don't like university bookstores. One semester I spent over $500 on books (okay I spend that much every semester but this semester made me mad) and I only got back 50 cents! FIFTY STUPID CENTS!
What was worse was that the fifty cents was from a book that I had forgotten to sell back the semester before.
I don't like people who try and rip off poor college students. Or poor people in general.
To this day that 50 cents is taped to my messageboard in remembrance of how much I don't like that place.

I hate random violence.
I hate reading in the newspaper that someone is beaten to death by a bunch of teenagers, because he comented on them blocking the sidewalk with their bicycles.
I hate reading that someone gets stabbed to death on a night out, because he had the guts to try to defend someone he didn't know when that person was being assaulted.
I hate hearing on the news that someone is shot, simply for having the 'wrong' skin color, wearing the 'wrong' clothes, saying the 'wong' things or walking/driving through the 'wrong' neighborhood.

I hate girls who are rude, bratty, have a "mightier-than-thou-art" attitude, just because they wear more expensive clohtes. In fact, I hate ALL people who act that way. It's not like I chose the family I was born into and wanted to be a "loser" in everybody elses eyes. I just AM.
Somehow, we all have the chance to come out of it. And when people refuse to see that you've changed? It's their problem, but it still hurts. If people would give others a second chance, like they'd want, maybe the world would be a better place. Not how it works. But just one person starting it could begin a whole chain reaction.
Now, onto my next rant.
Shoelaces. Why laces? Why don't we call it string? Shoestring. Has a nice ring to it. Or shoe yarn? I don't understand this. I could go on for hours about it! Shoelaces are the most useless invention ever, I think we should stick with velcro and buckles!
